Start Date:10/2002
Abstract:
Molecular systematics of fungi, particularly of the Ascomycota, relies heavily on the use of nuclear ribonsomal DNA. Use of these genes does not resolve all relationships with this phylum. This study aims to address these problems by elucidating the modes and rates of molecular evolution for 10 genes in taxa of the Sordariomycetes. Results can provide valuable information for the selection of alternatives to nuclear ribosomal DNA and lead to a more thorough understanding of evolutionary processes in the Ascomycota, and fungi in general.

Project Title:Aging aquatic animals using telomere length: influence of natural and anthropogenic stresses
Investigator(s):Dr. Leung K.M.Y.
Department:Ecology and Biodiversity
Source(s) of Funding:Block Grant Earmarked for Research
Start Date:10/2002
Completion Date:02/2004
Abstract:
To apply the quantitative polymerase chain reaction (q-PCR) technique for determining the telomere length in different tissues and blood samples of selected aquatic organisms (fish and mussels) with known ages, and construct a regression between age and telomere length for each species, under controlled laboratory conditions; to test the hypothesis: natural stresses (starvation or temperature) affect the telomere length; to test the hypothesis: chemicals (trace metals or hydrocarbons) affect the telomere length; use the established calibration curve to predict the age of the same species collected from wide population, based on their telomere length from the blood sample.

Project Title:Aquatic ecological risk assessment: comparison of tropical and temperate species sensitivity to chemicals
Investigator(s):Dr. Leung K.M.Y., Prof. Lam P.K.S.
Department:Ecology and Biodiversity
Source(s) of Funding:RGC Projects (Block Grant Funded)
Start Date:07/2003
Abstract:
To compare the species sensitivity distributions for temperate and tropical organisms exposed to individual chemicals, using existing databases, such as U.S. Environmental Protection Agency AQUIRE aquatic toxicology database, European ECETOC database, and toxicity data available in refereed literature and Governmental reports; to identify specific chemical groups, classified by mode of action or physico-chemical properties, for which technically sound estimates of tropical PNECs can be made/predicated on the basis of temperate toxicity data; to validate these predictions through the generation/compilation of extensive ecotoxicity datasets for a number of model substances; to establish a procedure to estimate tropical PNECs for the selected substances. Existing data will be supplemented by new eco-toxicity data where these are likely to result in improved confidence in the estimation of tropical PNECs.

Project Title:Ecological risk assessments of heavy metals and persistent organic pollutants on demersal fish and stomatopod species in the marine environment of Hong Kong
Investigator(s):Dr. Leung K.M.Y., Prof. Lam P.K.S.
Department:Ecology and Biodiversity
Source(s) of Funding:Block Grant Earmarked for Research
Start Date:12/2003
Abstract:
To determine the body burdens of common pollutants such as heavy metals, polycyclic aromatic hydrocarbons (PAHs) and organochlorines (OCs) in the common demersal fish and mantis shrimp species collected by bottom trawlers from various places in Hong Kong during the winter (dry) and summer (wet) months; to investigate the spatial and temporal differences in the body burdens of the above pollutants; to construct species sensitivity distributions (SSDs) for marine fish and crustaceans, respectively based on secondary chronic or acute eco-toxicity data [2,3]; to assess the ecological risks of the above pollutants on the target species using established probabilistic approaches [4,5].

To determine spatial variation in 16SrDNA-defined and morphological community structure, and phylogenetic relationships of microbial mat organisms among geographically distinct hot springs along gradients of latitude and altitude in China, Thailand and the Philippines; to establish temporal variation in 16SrDNA-defined and morphological community structure of microbial mat organisms in a selected hot spring location, with particular reference to tropical rainfall and UV incidence, and acute heat-shock events; to determine growth optima in vitro for selected hot spring cyanobaceria with regard to temperature and UV-irradiance; to examine the molecular basis of heat-and light-shock responses for selected hot spring cyanobacteria by quantitating expression of Hsp70 (dnaK) and nblS stress-induced genes during in vitro growth.

Investigator(s):Dr. Williams G.A., Dr. Leung K.M.Y.
Department:Ecology and Biodiversity
Source(s) of Funding:Block Grant Earmarked for Research
Start Date:11/2003
Abstract:
To optimize heat shock protein detection and quantification in different tissues of intertidal marine invertebrates (barnacles, mussels and limpets); to establish the in situ profile of heat shock protein (hsp) expression with relation to time and temperature; to explore the relevance of chaperone molecules in biogeographical distribution and habitat partitioning of intertidal organisms; to determine the cellular heat shock response of intertidal organisms under controlled laboratory experiments and the relevance of acclimatization in thermotolerance threshold; to test the hypothesis that organisms found at different tidal levels will exhibit different hsp expression profiles; to test the hypothesis that laboratory acclimatization will alter the thermotolerance threshold and hsp expression profiles of organisms.
