| It is imperative to have a better understanding of the mechanisms leading to various cataracts such that preventive measures can be developed. Since maintaining the ionic strength and ionic composition in the lens is essential for the transparency of the lens, we plan to study the mechanism of osmoregulation in this organ. Transgenic mice that over-express aldose reductase and sodium dependent myoinositol transporter in their lenses will be used to increase the lens' level of sorbitol and myoinositol, respectively. We will determine if high level of these osmolytes causes cataract development, and see when the level of one osmolyte is high, will the lens compensate by reducing the level of other osmolytes. We will also determine whether high intracellular osmotic pressure affects the regulation of expression of genes that control the level of osmolytes such as aldose reductase, sodium dependent myoinositol transporter, and taurine transporter. |

| Abstract: |
| Liver cancer is one of the major causes of cancer related mortality in this region. The only effective treatment is by surgical removal when discovered early. Chemotherapy is ineffective. We recently discovered that a large percentage of the human liver cancers overexpress aldose reductase (AR) and an aldose reductase-like gene (ARL-1). Since these two enzymes can act on a broad spectrum of substrates, it is likely that they can convert non-toxic chemicals to cytotoxic drugs that will specifically kill cells that overexpress them. We have identified naphthalene as a potential candidate for AR activated drug. In this project, we will test this concept in liver cell culture and in rat hepatoma model. This may lead to a more effective chemotherapy for liver cancers. |

| To develop transgenic mice that express aldose reductase (AR) specifically in the collecting tubules of the kidney to see if that would correct the defect in the urine concentrating ability in the AR deficient mice. The project will use the AR deficient mice to determine if AR contributes to hyperglycemia-induced oxidative stress, and to study its role in the detoxification of cytotoxic metabolites such as methylglyoxal and hydroxynonenal. |

| To elucidate the mechanisms of inhibition of MTP gene expression by insulin; to characterize the transcription factor(s) that activate the novel insulin negative response element present in the promoter of the MTP gene; to identify the physiological signals that regulate intestinal MTP gene expression using the human intestinal carcinoma Caco-2 cell culture system; to delineate the cause effect relationship between MTP level and lipoprotein production, in particular whether elevated MTP level influcences the subtype of lipoproteins produced; we have recently identified an oral hypolipidemic agent which inhibits MTP activity. We will determine the effects of this compound on body weight, plasma lipid profile, and glucose levels in the established Zucker obese diabetic rat model. Information gained from this research will provide insight into the molecular mechanisms and consequences of MTP regulation, which is of great importance in the management of hyperlipidemia and dyslipidemia. |

| To investigate the possible role of endothelin-1 (ET-1) in testicular blood flow and function. ET, a 21-amino acid peptide, is one of the most potent vasoconstrictor originally isolated from conditioned medium of cultured porcine aortic endothelial cells. |
